LATTOFERRINA

Lactoferrin, also known as lactotransferrin, is a multifunctional, iron-binding glycoprotein belonging to the transferrin family, originally identified in bovine milk and subsequently isolated from human and bovine milk. It is also found physiologically in the blood, as well as in some secretions (tears, semen, vaginal secretions, sweat). Lactoferrin plays different roles in innate immunity, and numerous studies have shown anticancer, antiviral, antifungal and antibacterial properties. For many years these activities have been attributed to the ability of lactoferrin to sequester iron thus depriving pathogens of this essential nutrient. However, it is now known that lactoferrin has a second type of antimicrobial activity, given by the result of the direct interaction between lactoferrin and some functional and structural components of pathogens and viruses: the antiviral action of lactoferrin is in fact given by its ability to bind to the glycosaminoglycans of cell membranes. In this way, it prevents the entry of viruses into the cell, guaranteeing a stop of the infection from the initial stages. This mechanism has proved effective against viruses: Herpes simplex, Cytomegalovirus, and influenza viruses.

Numerous studies have recently been carried out on the antiviral properties of lactoferrin including severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) which causes COVID-19

  • It has been experimentally observed that lactoferrin exerts a protective role in the host defense against SARS-CoV-2 infection through the binding of lactoferrin with HSPG (Heparan sulfate proteoglycan) present outside the cell plasma membrane, preventing thus the interaction between SARS-CoV-2 and host cells and hindering the entry of the virus into the cell. In addition, current knowledge suggests that COVID-19 mortality is not simply due to viral infection but is the result of a cytokine storm associated with hyper-inflammation: high levels have been found in severe cases of Covid-19. of interleukin 6 (IL-6), tumor necrosis factor alpha (TNFα) and ferritin. In this regard, lactoferrin has been shown to reduce IL-6 and TNFα and to act on the down-regulation of ferritin. Therefore, lactoferrin could be a candidate adjunct treatment for more severe cases of COVID-19.
  • Furthermore, lactoferrin has been shown to have a synergistic action with fluconazole against certain fungi, suggesting that it can potentially be used in combination against drug-resistant Candida infections. The synergistic action of lactoferrin with antibiotics, antifungals and antibacterial agents increases their effectiveness. Finally, lactoferrin also has interesting antioxidant properties. This glycoprotein acts as a modulator by donating iron when needed and withdrawing it when it is in excess, thus reducing the risk of formation of radical species and protecting cells from oxidation.

ZINC

Zinc is an essential trace element for living organisms and their biological processes, it is present in all tissues of the human body, especially at the intracellular level. Zinc is a cofactor for over 300 enzymes, including superoxide dismutase, alcohol dehydrogenase, and many proteases. Its action is important for maintaining the normal function of the immune system, in fact it is an essential element for its functioning by influencing the proliferation of cells of the immune system and therefore actively implementing the body's defense system from the aggression of pathogens . Zinc supplementation has also been proposed as a valuable aid to COVID-19 patients. In fact, many hospitalized patients with COVID-19 have been seen to have very low blood levels of zinc, which can contribute to the severity of the infection. In addition, zinc has been shown to have a powerful antiviral effect by influencing the metalloproteases involved in the coronavirus fusion process, decreasing both cell entry and cell-cell fusion responsible for virus propagation.
