Kit for the detection of antigens (based on colloidal gold technology) of Coronavirus 2 severe acute respiratory syndrome (SARS-CoV-2).
The double antibody sandwich method is adopted in the product and the measurements are carried out in solid phase with immunochromatographic principle.
The sample to be analyzed diffuses upwards at the end of the charger by capillary action, then the SARS-CoV-2 antigen in the sample binds with the antibody present in the marker buffer and forms an antigen-antibody-conjugated complex with gold colloidal.
This complex continues to diffuse with the sample up to the dinitrocellulose membrane and is blocked by the antibody-rich T line (test line), thereby forming an antibody-rich colloidal gold-labeled immune complex, rich in antibody.
The conjugate complex that has not been blocked continues to migrate upward and binds to the C line (quality control line), indicating that the aeration is complete.
The T line (test line) of the nitrocellulose membrane is coated with approximately 1.0 mg / mL of the SARS-CoV-2 monoclonal antibody, while the C line (quality control line) is coated with approximately 1.0 mg / mL of internal reference protein C.
How to use
Sample collection
- Nasal swab: the staff responsible for taking the sample must gently grasp the patient's head with one hand, hold the swab with the other, then insert it inside the nostril and continue along the floor of the nasal choana. nasal arch, pay attention to act gently to avoid traumatic bleeding. When the tip of the swab reaches the back of the pharyngo-nasal cavity, gently rotate the swab, making a full circle (in case of cough, pause for one minute), then slowly withdraw it and insert it into the tube with the eluent.
- Throat swab: to start, the patient needs to rinse their mouth with a saline solution; then the staff responsible for taking the sample must immerse the swab in the sterile saline solution (avoid immersing the swab in the virus preservation liquid and prevent the antibiotics causing allergies), tilt the patient's head slightly upwards, which must hold the mouth wide open, make "Ah" sound and show both sides of pharyngeal tonsils. The person in charge must use the swab to pass the tongue, rub out on both sides of the patient's tonsils at least 3 times, then rub on the back of the throat, up and down, and finally immerse the swab in the tube with the eluent.
Samples should be eluted within one hour of collection and then analyzed as soon as possible. If immediate processing is not possible, store samples under the following conditions: Sample can be stored for one day at 2 ° C to 8 ° C and long-term at -70 ° C or below.
Samples must be fully returned to room temperature before testing. Samples stored at low temperatures can only be used after they have been fully melted, heated and evenly mixed. Avoid freezing and thawing repeatedly.
Test methods
